The lab just got harder. Execute the perfect cloning pipeline, but beware of the new molecular traps! ✂️ 1. Digest: Cut the plasmid. 🧽 2. Phosphatase: Remove 5' phosphates. 🔥 3. Heat Kill (65°C): Denature the phosphatase! 🔗 4. Ligase: Add insert + enzyme + ATP. 🌡️ 5. Heat Shock (42°C): Open cell pores. ❄️ 6. Ice: Close pores to trap plasmid. 🚨 NEW EXAM TRAPS:
1. If you forget to Heat Kill the Phosphatase, it will destroy your insert!
2. You only have 2 SECONDS to hit ICE after Heat Shock, or the cells melt!
